Quality control tests
Ligature:
After a 2-fold over digestion with Abs I, ~ 90% of the DNA fragments can be ligated with T4 DNA ligase and re-cut.
16-hour incubation:
A 50 µl reaction containing 1 µg of pUC19SE / DriI and 2 units of enzyme incubated for 16 hours resulted in the same DNA banding pattern as a reaction incubated for 1 hour. A long incubation time can cause star activity.
No detectable degradation of a single or double-stranded oligonucleotide was observed after incubation with 1 unit of enzyme for 3 hours.
